What can you learn about leads, when you send them a marketing email?

What is possible to learn about a lead when they respond to your marketing email, besides when they are most likely to read email from you... Is it possible to determine if they forward your email around? Did they influence someone else to respond to your email? Are they a gatekeeper to meet with someone? Are they really interested in your product or service? What abour event invitations? Do they participate?
How can you tell what subjects they are interested in? What are the KPI's you look for in changing a lead to a customer and a customer into a brnad ambassador?
